Please help me, this has been doing my head in.

On the album Very Mercenary by The Herbaliser there is a sample at the beginning of the track, Who's the Realest. I'm sure it's Richard Pryor and Gene Wilder but cannot remember from which movie.
The quote goes:

G.W. This is real sh!t
R.P It's good sh!t, right?
G.W. I mean bad sh!t
R.P. Bad sh!t, like this sh!t is bad?
G.W. It's sh!t sh!t, this sh!t isn't worth sh!t. Anybody caught on the street with this wold get killed. This is real sh!t.

The dialogue is NOT from a Gene Wilder-Richard Pryor movie.

Instead, it's from "Running Scared" (1986), which starred Billy Crystal and the late Gregory Hines, who passed away last month.

The dialogue exchange you're talking about is with Hines and a laboratory technician played by Jim Ortlieb, a longtime character actor.
